The thiol 2-chlorotrityl resin is an acid-labile resin for the preparation of symmetrical disulfides. We can use the method of alkylation or acylation of the polymer-supported thiol group to formate an immobilized thiol [1] or thioacid, which can then be released by treatment with 50-95% TFA in DCM containing 5% TIS. It is very difficult to substitute at the highly hindered trityl thiol, so long reaction times or highly activated reagents may be required to effect complete reaction. Using an excess of Fmoc-amino acid activated with an equivalent amount of Melm in dry DCM can best effect the acylation with an amino acid. The symmetrical disulfides can be released by treatment with electrophilic oxidants such as I2 or TI3+.
